Anti – Gout.

 

COMPOSITION:-

Each tablet contains:- Allopurinol 200 mg

INDICATIONS:-

Allopurinol is used to reduce urate concentrations in body fluids and/or urine to prevent or reverse the deposition of urate/uric acid.
Allopurinol is primarily indicated for the main clinical manifestations of urate/uric acid deposition. These are gouty arthritis, skin tophi and/or renal involvement through crystal-deposition or stone formation.
Such clinical manifestations occur in:
- idiopathic gout.
- uric acid lithiasis.
- acute uric acid nephropathy.
- neoplastic disease and, myeloproliferative disease with high cell turnover rates, in which high urate levels occur either spontaneously or after cytotoxic therapy.

    - certain enzyme disorders which lead to overproduction of urate and involve:-
hypoxanthine guanine phosphoribosyltransferase including Lesch-Nyhan syndrome.
glucose-6-phosphatase including glycogeri storage disease.
phosphoribosylpycophosphate synthetase.
phosphoribqsylpyrophosphate amidotransferase.
adeninephosphoribosyltransferase.
glutathionereductase.
glutamate dehydrogenase.
Allopurinol is indicated for the prevention and treatment of calcium oxalate / phosphate renal stones in the presence of hypemricaemia and/or hyperuricosuria.
Allopurinol may be indicated for clinical manifestations of urate deposition or hyperuricaemia and/or hyperuricosuria of varying significance, which may be associated with:-
certain acquired or congenital renal anomalies.
certain disorders of metabolism.
certain endocrine-related disorders.
haemolytic anaemias.
megaloblastic-anaemia.
administration of certain drugs.
certain other miscellaneous conditions.
   
DOSE:-

Adults:

2 to 10 mg/kg bodyweight/day, or
100 to 200 mg daily in mild conditions.
300 to 600 mg daily in moderately severe conditions
700 to 900 mg daily in severe conditions.

Children under 15 years:

10 to 20 mg/kg bodyweight/day or
100 to 400 mg daily
Use in children is rarely indicated except in malignant conditions (especially leukaemia) and certain enzyme disorders, particularly Lesch-Nyhan syndrome.

The dose should be titrated against the patient by monitoring serum urate concentrations and urinary urate/uric acid levels at appropriate intervals.
